Semax vs Selank: Comparing Two Nootropic Peptides

Semax and Selank get mentioned together so often that people assume they are interchangeable. They are not. Both came out of the Russian Academy of Sciences, both are heptapeptides, and both have decades of clinical use behind them in Russia. But they were built to solve different problems, they work through different mechanisms, and they feel different to the people who use them.

This article breaks down what separates the two, where they overlap, and why a lot of researchers end up running them together rather than picking one.

Where They Came From

Both peptides are products of the same research lineage, which is part of why they pair so naturally.

Selank is a synthetic analog of tuftsin, a naturally occurring tetrapeptide (Thr-Lys-Pro-Arg) found in the heavy chain of human immunoglobulin G. Researchers added a Pro-Gly-Pro tripeptide to the C-terminus, which made the molecule metabolically stable enough to cross the blood-brain barrier and act on the central nervous system. It was developed specifically as an alternative to benzodiazepines and has been a prescription anxiolytic in Russia since the early 2000s.

Semax is a synthetic fragment of adrenocorticotropic hormone, based on the ACTH(4-10) sequence that carries the cognitive effects of the hormone, modified for stability and brain penetration. The name comes from the Russian for “seven amino acids.” It has been used clinically in Russia for decades and sits on Russia’s List of Vital and Essential Drugs.

The shared origin matters less than the shared design philosophy. Both were engineered to deliver a clinical effect through endogenous regulatory pathways rather than brute-force receptor activation. That is why neither one produces the crash, the dependence, or the cognitive dulling you see with the conventional drugs they were designed to replace.

How They Work

This is where the two peptides genuinely diverge.

Selank: GABA Without the Sedation

Selank’s headline mechanism is positive allosteric modulation of the GABAa receptor. GABA is the brain’s main inhibitory neurotransmitter. Benzodiazepines, barbiturates, and alcohol all hit this same system, but they do it by directly activating the receptor, which is what produces sedation, amnesia, tolerance, and dependence.

Selank does something more subtle. Instead of binding the primary site, it appears to bind a related but distinct allosteric site, shifting the receptor’s conformation so that GABA itself binds more effectively. You get enhanced inhibitory signaling without the receptor being forced open. Radioligand work (Kasian et al., 2019) showed Selank can compete with diazepam for binding on the receptor complex while producing functionally different downstream effects, which is the biochemical fingerprint of an allosteric modulator rather than a direct agonist.

That is not its only pathway. Selank also:

Semax: Building Capacity Through BDNF

Semax is essentially a BDNF lever. Brain-derived neurotrophic factor supports neuron survival, drives the growth of new neurons, and strengthens synaptic connections. More BDNF correlates with better learning and memory consolidation.

A single dose of Semax has been shown to raise BDNF protein by roughly 1.4-fold and BDNF mRNA by up to 3-fold, while increasing TrkB receptor phosphorylation 1.6-fold (Dolotov et al., 2006). TrkB is the receptor BDNF signals through, so Semax is not just producing more of the growth factor, it is amplifying the brain’s ability to respond to it.

The contrast is clean. Selank quiets an overactive stress system. Semax builds the underlying hardware. One is regulatory, the other is constructive.

What the Evidence Actually Shows

Both peptides have more human data behind them than most research compounds, which is worth saying plainly. The bulk of it comes from Russian clinical literature. That is not a reason to dismiss it, but it is a reason to read it with the same scrutiny you would apply to any single-country research base: the findings are consistent across multiple groups and models, and the absence of a Western regulatory file reflects which markets pursued approval, not a verdict on the data.

The pattern is telling. Selank’s best evidence is in anxiety. Semax’s best evidence is in cognition and neurological recovery. The research tracks the intended use of each peptide.

Stacking Them Together

This is the part most people are actually building toward. Selank and Semax are one of the most established peptide stacks in both research and clinical settings, precisely because their profiles are complementary rather than redundant.

Semax sharpens focus and drive. Selank takes the edge off without dulling anything. Run together, the reported result is calm focus: the productive alertness of Semax with the anxiety floor of Selank, minus the jitter or the sedation you would get trying to engineer the same state with stimulants and benzodiazepines.

They work through entirely different mechanisms (BDNF and dopamine versus GABA modulation), which is why they are generally described as having no meaningful interaction concerns. As with any stack, the conservative approach is to introduce one at a time so you know which compound is producing which effect before combining them.

The one Semax-specific flag worth highlighting is glucose. Diabetic users should monitor blood glucose closely. Semax also carries cautions around active cancer (BDNF can theoretically support tumor growth), seizure history, and bipolar disorder, all of which trace back to its neurotrophic and mood-pathway activity. Selank’s main cautions involve combining with other GABAergic compounds, serotonergic medications, and autoimmune conditions given its immunomodulatory action.

Neither has been studied in pregnancy or breastfeeding, so both should be avoided in those populations. And neither is a substitute for proper sleep, nutrition, stress management, or professional evaluation of an underlying condition.

Which One Should You Use?

The honest answer is that the question is usually framed wrong. People ask “Semax or Selank” as if they are competing for the same slot. They are not.

Selank is the calm. Semax is the sharp. The reason they are mentioned in the same breath so often is not that they are alternatives, it is that together they cover the full range of “I want to think clearly and not feel stressed while I do it.”

Is one stronger than the other? They are not on the same scale. Selank is the stronger anxiolytic, Semax is the stronger nootropic. Comparing their raw “strength” is like comparing a stronger painkiller to a stronger stimulant.
